I've tried to collect all the information I have on this in one place, the Dashlaunch plugin seems to be a working solution for all games but there are other methods which still work for some titles.
To start with,
Combining extracted files into a \games\gametitle folder
Castlevania: Lords of Shadow Files Data00.packed and Data01.packed from DVD2 must be renamed to Data02.packed and Data03.packed before merging folders to prevent overwriting files
Dead Space 2 Doesn't work
Final Fantasy XIII Note - most people report that overwriting DVD3's contents with DVD2's followed by DVD1's files produces a working merged folder.
Mass Effect 2 Most people say to copy DVD1's contents over the top of DVD2 but it shouldn't make a difference
Star Ocean: The Last Hope Doesn't work
Using the above method then converting the merged folder to an ISO then a GOD

Confirmed Mass Effect 2. I just made a 9+GB GOD after extracting discs, copying D1 over D2, ISO with createisogood, then GOD with ISO2GOD. It's working fine. It transfers over FTP or USB much faster than 3,000 files.

Thanks for posting that, Unfortunately this forum doesn't allow editing on posts after some time has passed so I can't edit it into the OP, Kinda disappointing. Also I didn't list a few games that come on two discs that aren't 'true' multidisc games such as Forza 3 - which come with a second disc which has only DLC.
These can all be copied from the content folder on the disc to the hard drive and then the first disc used normally.

Software I've used is probarbly not correct. I was using "Create Iso by Froggy_007 PreRelease" and ISO2GOD 1.3.5. When I attempted this the resulting joined GOD files showed ok in NXE (achievements on the right side like other GODs) but when launching I got the 'cannot start this game' errors.

I have the same problem as apopberz.
I copied the disk one content of Mass Effect 2 over the disk 2. Then I created the iso with exiso.exe since all the create_iso I found here had a trojan in it. So I got the exiso from xbox hacker and used that to create the iso from the compined disk one disk two folde. And then I proceeded to ISO2GOD(1.3.5). Now i get 'cannot start this game' error.
ska_drummer85 can you confirm the versions of the softwares you used when to do this and if you are launching your game in the kinect dash.

I had the same problem with ME2, seemed like extracting both to same dir then making a shortcut with quickboot was the only way it would work (which isn't ideal since achievements don't show up on the side), but I found a solution now by converting the first disc to GOD, extracting the second disc, and then using Disk Swapper to make it work, plays like a charm now... It does seem to take a second or so longer to switch between discs than with both discs extracted, but that's really not a big deal since disc swaps don't come up that often anyway (sure beats swapping the physical discs every time though!)

I've had success myself with the merged folder method with ME2 - launching from FSD. but taking that same, working folder and using the iso then god tools I mentioned gives the error. ska_drummer85 Needs to get back to us with the tool they used for the iso creation and then I/we can test other titles. All my games work from merged folders apart from DS2 (I haven't tested but I'd bet using the castlevania method works) but I haven't got a single one working as a merged GOD yet
