I dont think so. You only have to have the XNA Runtime enviroinment, which is free. You can download it from Live Marketplace. If you want to make your own games and deploy them on the xbox 360, then you would have to pay the annual ~100$ fee.

if any of you guys used ur analytical skills at all you'll realise the point of the announcement was that the game will be open to everyone to purchase on xbla! Not to play on a xna subscription!
